Lac-Phe: Exercise-Induced Molecule Shows Potential for Weight Loss Therapies

A new study by researchers from Baylor College of Medicine, the Duncan Neurological Research Institute, Stanford University, and collaborating centers has revealed how exercise-induced Lac-Phe suppresses appetite and supports weight loss. Published in Nature Metabolism, the findings show that Lac-Phe, a metabolite that rises sharply after intense activity, directly influences brain circuits regulating hunger.

In mice, Lac-Phe inhibited AgRP neurons—cells that drive hunger—thereby activating PVH neurons, which reduce food intake. This mechanism curbed appetite without affecting normal behavior. Researchers also identified that Lac-Phe works by activating KATP channels on AgRP neurons; blocking these channels eliminated its appetite-suppressing effects.

Previous work demonstrated that Lac-Phe reduced food intake and promoted weight loss in obese mice without harmful side effects. While the current research focuses on animal models, the results highlight a promising pathway for developing weight management therapies that mimic exercise’s natural effects on appetite and metabolism.
