Pfizer’s oral CDK4 inhibitor atirmociclib has met its primary endpoint in a Phase II trial for advanced breast cancer. In the FOURLIGHT-1 study, the drug—combined with Faslodex (fulvestrant)—significantly improved progression-free survival compared to standard treatment options.
The trial included patients with hormone receptor-positive, HER2-negative metastatic breast cancer whose disease had progressed after prior CDK4/6 inhibitor therapy. Results were consistent across key patient subgroups, highlighting the drug’s potential effectiveness in a difficult-to-treat population.
While overall survival data are still immature, atirmociclib demonstrated a manageable safety profile, with low discontinuation rates due to side effects.
These findings support further development, including an ongoing Phase III trial in first-line treatment. However, competition remains strong, with alternative therapies and emerging drugs in the same class. Success in earlier treatment settings will be critical for atirmociclib’s future positioning in breast cancer care.
