Pfizer Eyes Earlier Use of Elrexfio After Strong Phase III Results in Multiple Myeloma

Pfizer is advancing plans to expand the use of its drug Elrexfio (elranatamab) into earlier treatment settings for relapsed or refractory multiple myeloma (RRMM), following positive Phase III trial results.

In the MagnetisMM-5 study, Elrexfio as a monotherapy delivered a statistically significant improvement in progression-free survival (PFS) compared to a standard-of-care regimen combining daratumumab, pomalidomide, and dexamethasone. The trial, which enrolled around 944 patients who had received at least one prior therapy, met its primary endpoint, with many patients remaining progression-free at interim analysis. Overall survival (OS), a key secondary endpoint, is still being evaluated.

Elrexfio demonstrated a safety profile consistent with previous findings. According to Pfizer, the results support its potential to benefit patients earlier in their treatment journey.

The company plans to engage with global regulators, including the U.S. Food and Drug Administration, to discuss next steps. Elrexfio is already approved in multiple markets for heavily pretreated patients.
