Mapping Genetic Activity Offers New Pathways for Personalized Cancer Treatment

City of Hope®, a leading U.S. cancer center, has co-led a study showing how genetic activity near chromosomes can reveal how cancer-causing mutations reshape DNA and the tumor environment. The research focuses on extrachromosomal DNA (ecDNA)—tiny DNA circles outside chromosomes—which drive aggressive tumor growth by promoting genetic instability and oncogene amplification.

Using spatial transcriptomics and genomic sequencing on glioma samples, researchers identified tumor cell clusters with shared origins and mutations. These insights enhance understanding of tumor evolution and may guide precision therapies.

The study also found ecDNA’s ability to adapt to environmental changes, including therapies, allows tumors to reprogram and persist. This discovery supports City of Hope’s goal to develop targeted, less toxic treatments and move closer to curing more cancers through personalized medicine.
